| 目的 探讨脂肪干细胞（（adipose-derived sterm cells,ADSCs ）覆膜聚丙烯补片能否减轻疝修补术后的炎症反应。 方法 培养扩增检测脂肪干细胞并接种到聚丙烯补片上，体外培养10天。分别将ADSCs覆膜聚丙烯补片和聚丙烯补片植入覆膜组和未覆膜组大鼠的腹壁肌肉表面，检测术前及术后3、7、14、21、28 天血常规、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的变化。 结果 ADSCs在聚丙烯补片上附着生长良好，免疫荧光检测显示干细胞特异性标记CD90表达≧97%。覆膜组在术后14d炎症反应高峰时期的白细胞计数［（10.856±0.556）vs （11.915±0.561）， P <0.05］、TNF-α［（87.12±12.59）vs （175.28±34.37）， P <0.01］、IL-1β［（275.69±54.28）vs （501.36±98.15）， P <0.01］均低于未覆膜组，有统计学意义。 结论 ADSCs覆膜聚丙烯补片相较聚丙烯补片植入大鼠体内后可以减轻全身炎症反应。 |

| [Abstract] Objective To investigate the effect of adipose-derived stom cells (ADSCs) coated polypropylene patch on relieving the inflammation after herniorrhaphy.Methods The adipose-derived stem cells were seeded into polypropylene mesh。In vitro culture for 10 days.The ADSCs covered with PP and PP were implanted into the abdominal wall muscle of the experimental group and the control group respectively. The blood samples were collected at 3, 7, 14, 21 and 28 days after operation.WBC count,TNF-α and IL-1β were measured.Results ADSCs grew well on the polypropylene patch, and the CD90 expression was more than 97% by immunofluorescence. In the peak period of inflammatory reaction at the 14 days in the experimental group , the WBC count[(10.856 ± 0.556) vs (11.915 ± 0.561), P <0.05], TNF-α [(87.12 ± 12.59) vs (175.28 ± 34.37), P <0.01], IL-1β (275.69 ± 54.28) vs (501.36 ± 98.15), P <0.01] is lower than that of the control group.Conclusion Compared with the polypropylene patch implantation Mouse body can reduce the systemic inflammatory response. |
