中文摘要:
      目的 研究仙鹤草水提液对不同肺癌细胞(HCC827、H358、H1650、H1299)的增殖抑制作用。方法 采用SRB法检测不同浓度仙鹤草水提液(0.4mg/ml、0.8mg/ml、1.6mg/ml、3.2mg/ml)分别作用24h、48h、72h对四种肺癌细胞株的抑制率的影响,并检测仙鹤草水提液与顺铂不同浓度联合作用72h对H1299的抑制率。结果 仙鹤草水提液对4种肿瘤细胞均有不同程度的抑制作用。作用72h对比,3.2mg/ml仙鹤草水提液对HCC827、H358、H1650的增殖抑制率与5μmol/L的顺铂相比无明显差异(P>0.05);而3.2m/ml的仙鹤草水提液对H1299的抑制效果明显优于5μmol/L的顺铂(84.74% vs 34.47%,P<0.05)。0.5μmol/L 顺铂与四个梯度浓度(0.4,0.8,1.6,3.2 mg/ml)的仙鹤草水提液对肺癌细胞H1299的抑制作用逐渐增加;与0.8mg/ml仙鹤草水提液联合用药对H1299的抑制作用(32.24%)低于单用该浓度的仙鹤草水提液(35.47%),高于单用该浓度的顺铂(12.81%);其他组合抑制作用均高于单用该浓度的仙鹤草水提液或顺铂,其中0.5μmol/L顺铂和3.2mg/ml仙鹤草水提液联合用药对肺癌细胞的增殖抑制作用强,抑制率达到92.49%。1.6mg/ml的仙鹤草水提液与四个梯度浓度(0.5,1,2.5,5μmol/L)的顺铂联合用药对肺癌细胞的增殖抑制作用均明显强于单用该浓度的仙鹤草水提液或顺铂,其中5μmol/L顺铂和1.6 mg/ml仙鹤草水提液联合用药抑制作用强,抑制率达到91.77%。结论 本实验表明仙鹤草水提液对肺癌细胞有显著的体外增殖抑制作用,并且联合顺铂用药产生协同作用,抑制作用强于单独用药。
英文摘要:
      Objective To investigate the inhibitory effect of agrimoniae water extraction on lung cancer cell line growth. Method Different concentration of agrimoniae water extraction(0.4mg/ml、0.8mg/ml、1.6mg/ml、3.2mg/ml)influence the lung cancer cell lines growth after effect 24h、48h、72h with SRB, and also study the effect of agrimoniae water extraction use with cisplatine after 72h. Results The grimoniae water extraction has inhibitory effect on the 4 kinds of lung cancer cells in?different?degree. Compared with cisplatin(5 μ mol/L) the proliferation inhibition rate of grimoniae water extraction (3.2mg/ml) on HCC827、H358、H1650 for 72h was not significantly different(P > 0.05) . However, the proliferation inhibition rate of grimoniae water extraction (3.2mg/ml) on H1299 is obviously higher than that of cisplatin(5 μ mol/L)(84.74% vs 34.47%,P<0.05). The cisplatin (0.5mol/L )with different concentration of agrimoniae water extraction (0.4, 0.8, 1.6, 3.2 mg/ml) has the gradually increased inhibitory effects on lung cancer H1299 cells; The inhibitory effect of combination with 0.8mg/ml (32.24%) on H1299 is lower than that of single agrimoniae water extraction with the same concentration (35.47%) and higher than that of single cisplatin with the same concentration (12.81% ).The inhibitory effect of other combinations was higher than that of the single agrimoniae water extraction or cisplatin and The inhibitory effect of 3.2mg/ml agrimoniae water extraction combined with 0.5 mol/L cisplatin is strongest, reached 92.49%. The inhibitory effect of 1.6mg/ml agrimoniae water extraction combined with four concentration of cisplatin (0.5, 1, 2.5, 5 mol/L) on lung cancer cell was significantly stronger than single agrimoniae water extraction or cisplatin and the inhibitory effect of 5μmol/L cisplatin combined with 1.6mg/ml agrimoniae water extraction is strongest, reached 91.77%. Conclusion The experimental results infer that agrimonia is a much efficient anti-tumor potential drug worthy keep studying.
