      目的 探讨过坛龙提取物对四氯化碳 ( CCl4 )致小鼠急性肝损伤小鼠的保护作用及其机制。方法:腹腔注射四氯化碳诱导小鼠急性肝损伤模型,将50只小鼠随机分为5组,每组10只,设置空白对照组、模型组、过坛龙提取物高、低剂量组、阳性对照组(联苯双酯混悬液),空白对照组和模型对照组。测定小鼠血清谷丙转氨酶 ( ALT )、谷草转氨酶 ( AST )、肝组织中丙二醛 ( MDA )、一氧化氮(NO) 含量,一氧化氮合酶(NOS)的活性,光镜观察肝脏病理学改变。结果:过坛龙提取物低、 高剂量组小鼠谷丙转氨酶 (ALT)、 谷草转氨酶 (AST) 、丙二醛 ( MDA )、一氧化氮(NO)、一氧化氮合酶(NOS)等较模型组均有显著降低[ALT:(365.4±123.5)U/L、(275.1±21.9) U/L 比(443.2±227.3) U/L,P<0.05, P<0.01; AST:(232.4±54.1)U/L、(195.9±11.8)U/L 比(297.9±132.1)U/L,P<0.05,P<0.01;MDA:(0.416±0.024)nmol/ml、(0.415±0.052)nmol/ml比(0.426±0.042)nmol/ml P<0.05,P<0.01;NO:(0.148±0.010)μmol/g、(0.141±0.015)μmol/g比(0.148±0.017)μmol/g P<0.05, P<0.01;NOS:(0.154±0.013)U/mg、(0.122±0.012) U/mg比(0.187±0.013)U/mg P<0.05, P<0.01]。表明过坛龙提取物可明显降低CCl4所致肝损伤小鼠的血清ALT和AST活性,减少丙二醛(MDA)和NO的含量,降低NOS活性;病理学检查显示,过坛龙提取物各剂量组较模型组肝细胞肿胀、变性、坏死程度减轻,其中高剂量组改善更显著。结论:过坛龙提取物对急性肝损伤小鼠具有保护作用。

      ABSTRACT Objective to To investigate the protective effects and the related mechanisms of extract from flabelate maidenhair herb against acute liver injury induced by carbon tetrachloride ( CCl4) in mice.Methods the models of acute liver injury were induced by Intraperitoneal injection CCl4 in rats. Fifty ICR mice were randomly divided into five groups(n= 10 in each ): normal control group, model group, bifendate pills group,extract from Flabelate maidenhair herb groups(high dose and low dose). The serum activities of alanine aminotransferace (ATL),aspartate aminotransferase (AST) were determined , the liver index,the contents of malonaldehyde ( MDA) ,the concentration of NO and the activity of NOS in liver tissue of each group were measured. pathological changes of liver tissue were examined under light microscope. Results Flabelate maidenhair herb groups showed decreased, ALT, AST, MDA, NO, and NOS compared with model group(ALT:365.4±123.5U/L,275.1±21.9U/L vs 443.2±227.3U/L; AST: 232.4±54.1 U/L,195.9±11.8U/L vs 297.9±132.1U/L;MDA:0.416±0.024 nmol/ml,0.415 ±0.052nmol/ml vs 0.426±0.042 nmol/ml;NO:0.148±0.010μmol/,0.141±0.015μmol/g vs 0.148±0.017μmol/g;NOS:0.154±0.013U/mg、0.122±0.012 U/mg vs 0.187 ±0.013U/mg P<0.05,all P<0.05).Compared with CCl4 model group,The serum activities of ATL and AST, the contents of MDA,the concentration of NO and the activity of NOS in liver tissue were obviously decrdased after preventive treatment with extract from Flabelate maidenhair herb. Compared with model group ,The pathological examination in both high and low dose groups showed liver cellular swelling,degeneration and necrosis relieved,and pathological injury had been improved, especially in high -dose group .Conclusion extract from Flabelate maidenhair herb has a protective effect on acute liver njury induced by CCl4 in rats. This might be associated with the anti-lipid-peroxidation effect and decrease NOS activities NO levels.
