| 目的：建立寒凝血瘀证荷瘤小鼠复合动物模型，观察寒凝血瘀状态及温阳活血法对肿瘤转移靶组织微环境的影响。方法：将48只Balb/c小鼠随机分为空白对照组、单纯荷瘤组、寒凝血瘀荷瘤组、温阳活血治疗组分别造模，每组12只，Western blot方法检测各组小鼠肺组织中MMPs及ERK的表达。结果：与单纯荷瘤小鼠检测结果相比较，寒凝血瘀荷瘤组小鼠ERK1/2、MMP-2表达均升高，差异有统计学意义[ERK1/2 RI值：(1.40±0.01)比(0.98±0.03)，P<0.05]；[MMP-2 RI值：（1.26±0.05)比（1.00±0.04)，P<0.05]；与寒凝血瘀荷瘤组小鼠检测结果相比较，温阳活血治疗组小鼠ERK1/2、MMP-2表达均降低，差异有统计学意义，[ERK1/2 RI值：(1.18±0.01)比(1.40±0.01)，P<0.05]；[MMP-2 RI值：(1.01±0.05)比(1.26±0.05)，P<0.01]。与寒凝血瘀荷瘤组小鼠比较，温阳活血治疗组小鼠MMP-12的表达升高，差异具有统计学意义，[MMP-12 RI值：(1.23±0.03)比(1.08±0.06)（p＜0.05）]。
结论：温阳活血法通过下调荷瘤小鼠肿瘤转移靶器官肺组织ERK1/2、MMP-2的表达及上调MMP-12从而影响肿瘤基质及肿瘤血管生成，进而减少转移灶形成的发生率。 |

| Objective: To establish a mouse model of colorectal carcinoma with cold coagulation and blood stasis, given Yang –warming and blood-activating therapy as the treatment. Then we explore its role in tumor metastasis and the effects of Yang –warming and blood-activating therapy though the detection of MMP-2、MMP-12、ERK1/2 in mice. Methods:48 Balb/c mice were randomly divided into 4 groups,respectively called blank control group、tumor-bearing group 、cold coagulation and blood stasis tumor-bearing group、Yang –warming and blood-activating therapy group.Parts of the lung tissue was tested the expression of MMPs and ERK by Western blot . Results: Compared with the tumor-bearing mice group, ERK1/2、MMP-2 of the cold coagulation and blood stasis tumor-bearing group was greater[ERK1/2 RI值：(1.40±0.01)比(0.98±0.03)，P<0.05]；[MMP-2 RI值：（1.26±0.05)比（1.00±0.04)，P<0.05]；Compared with the cold coagulation and blood stasis tumor-bearing group，ERK1/2、MMP-2 of the Yang –warming and blood-activating therapy group was reduced[ERK1/2 RI值：(1.18±0.01)比(1.40±0.01)，P<0.05]；[MMP-2 RI值：(1.01±0.05)比(1.26±0.05)，P<0.01]。Compared with the cold coagulation and blood stasis tumor-bearing group，MMP-12 of the Yang –warming and blood-activating therapy group was greater[MMP-12 RI值：(1.23±0.03)比(1.08±0.06)（p＜0.05）]。Conclusions: Yang –warming and blood-activating therapy can reduce the incidence of tumor metastasis in different degrees. |
