目的:研究自噬在丹参酮IIA减轻内毒素性急性肺损伤中的作用及相关机制。方法:将32只SD雄性大鼠随机分为四组:正常对照组、模型组、丹参酮IIA组及3-MA组(自噬抑制组)。通过气管内滴入内毒素( LPS) 建立大鼠急性肺损伤模型及腹腔预注射自噬抑制剂3-MA或丹参酮IIA后提取肺组织,免疫组化观察肺组织的病理改变,Elisa试剂盒检测炎症因子TNF-α、IL-6、IL-10水平,western blotting检测自噬相关蛋白LC3-Ⅱ及泛素相关蛋白SQSTM1/P62水平。结果:免疫组化示模型组肺泡间隔增厚,肺泡腔狭窄,大量炎症细胞渗出;丹参酮IIA组则明显改善;3-MA组抑制自噬后肺泡炎症细胞渗出明显。与对照组相比,内毒素(LPS)诱导的急性肺损伤组自噬水平增强,炎症因子浓度升高;丹参酮IIA组自噬水平进一步增强,炎症因子浓度降低;而3-MA组自噬水平降低,炎症因子浓度升高。结论:丹参酮IIA能增强内毒素性急性肺损伤中的自噬水平,增强的自噬水平在丹参酮IIA减轻内毒素性急性肺损伤中具有保护作用。 |

Aims:To investigate the effect and related mechanisms of autophagy in Tanshinone IIA reducing endotoxin induced acute lung injury.Methods:Thirty-tworabbits were divided randomly into four groups(n=8):control group、acute lung injury model group、Tanshinone IIA group and autophagy inhibition group.Acute lung injury model group was given lipopolysaccharide intratracheally establish acute lung injury model,throughpretreatment with intraperitoneal injectionautophagy inhibitor 3-MA or Tanshinone IIA after establish acute lung injury model, extract the lung tissue and the expression of inflammatory factors were detected by immunohistochemistry method, the levels of inflammatory cytokinesTNF-α、IL-6、IL-10were measured with Elisa kits,autophagy related protein LC3-II and P62 /SQSTM1 were measured with Western blotting.Results:Histopathologic examination show acute lung injury model groupedematous changes of alveolar walls, massive polymorphonuclear infiltration were observed, however,Tanshinone IIA group significantly alleviated while autophagy inhibition group damage was observed. Compared with control group,the level ofautophagyincreased in endotoxin inducedacute lung injury groupandinflammatory cytokineslevelsrised, and the level of autophagyfurther increased in Tanshinone IIA group while inflammatory cytokines levelsdecreased, however, the level of autophagy in autophagy inhibition group was decreased, inflammatory cytokineslevels wereincreaesed.Conclusions: Tanshinone IIA can enhance the level of autophagy in endotoxinreducdacute lung injury.Enhanced the levelof autophagy plays a protective rolein tanshinone IIA reducing endotoxininducedacute lung injury. |
