      目的 明确加味麻杏石甘汤对大鼠急性放射性肺炎的干预作用,进一步探讨其作用机理,为加味麻杏石甘汤在临床应用上提供指导性意义。 方法 复制急性放射性肺炎SD大鼠模型,按中药高、中、低剂量组分别给予加味麻杏石甘汤灌胃,阴性对照组、模型组予生理盐水灌胃,2ml?220g-1?d-1,连续8周。分别于第2、4、8周各组随机处死4只大鼠,取肺组织,采用Western-Blot方法检测大鼠肺组织中P-Smad2、P-Smad3、Smad6、Smad7蛋白的表达情况。 结果 经照射后,模型组大鼠P-Smad2蛋白(0.256±0.003)较阴型对照组(0.194±0.004)表达升高(P﹤0.01),中药高、中、低剂量组P-Smad2表达(0.098±0.011、0.106±0.007、0.105±0.006)均较模型组(0.256±0.003)下降(P均﹤0.01);经照射后,模型组大鼠P-Smad3蛋白(1.253±0.005)较阴型对照组(0.506±0.006)表达升高(P﹤0.01),中药高、中、低剂量组P-Smad3(0.666±0.005,0.536±0.004、0.919±0.007)均较模型组(1.253±0.005)下降(P均﹤0.01);照射后,模型组大鼠Smad6表达(0.164±0.075)较阴型对照组(0.468±0.191)下降(P﹤0.01),中药中、低剂量组Smad6(0.283±0.014、0.337±0.029)较模型组(0.164±0.075)上调(P均﹤0.01);照射后,模型组大鼠Smad7表达(3.674±0.626)较阴型对照组(4.586±0.287)下降(P﹥0.01),中药高、中、低剂量组Smad7(5.565±1.170、9.631±0.566、8.525±0.50)较模型组(3.674±0.626)上调(P均﹤0.01)。 结论 ①经照射后,大鼠肺组织中P-Smad2、P-Smad3水平均上升,Smad6、Smad7水平均下降。②加味麻杏石甘汤能下调急性放射性肺炎大鼠肺组织中P-Smad2、P-Smad3蛋白的表达,能上调急性放射性肺炎大鼠肺组织中Smad6、Smad7蛋白的表达。

      Objective Determining the intervention of Modified Maxing Shigan Decoction on acute radiation pneumonitis in rats and exploring the mechanism further was for providing guiding significance of the clinical applications of Modified Maxing Shigan Decoction. Methods Replicated model SD rats with acute radiation pneumonitis, the Modified Maxing Shigan Decoction groups(high-dose group, middle-dose group, low-dose group) were given intragastric administration with Modified Maxing Shigan Decoction 2ml?220g-1?d-1 8 weeks. The model group and the negative control group were given intragastric administration with normal saline(2ml?220g-1?d-1) 8 weeks. Respectively in the 2nd week, 4th week and 8th week, 4, 4 and 4 rats of every groups were sacrificed randomly, and took out the lung tissues. The expression of P-Smad2、P-Smad3、Smad6、Smad7 protein were analyzed by Western-Blot technique. Results After the radiation, the expression of P-Smad2 protein of the model group were up-regulated as compared with the negative control group (0.256±0.003 vs 0.194±0.004, P﹤0.01). After treatment the expression of P-Smad2 protein of the high-dose group, middle-dose group and low-dose group were lower than that in the model group (0.098±0.011、0.106±0.007、0.105±0.006 vs 0.256±0.003, all P﹤0.01). The expression of P-Smad3 protein of the model group were up-regulated as compared with the negative control group (1.253±0.005 vs 0.506±0.006, P﹤0.01). The expression of P-Smad3 protein of the high-dose group, middle-dose group and low-dose group were lower than that in the model group (0.666±0.005,0.536±0.004、0.919±0.007 vs 1.253±0.005, all P﹤0.01). After the radiation, the expression of Smad6 protein of the model group were down-regulated as compared with the negative control group (0.164±0.075 vs 0.468±0.191, P﹤0.01). After treatment the expression of Smad6 protein of the middle-dose group and low-dose group were higher than that in the model group (0.283±0.014、0.337±0.029 vs 0.164±0.075, all P﹤0.01). The expression of Smad7 protein of the model group were down-regulated as compared with the negative control group (3.674±0.626 vs 4.586±0.287, P﹥0.01). After treatment the expression of Smad7 protein of the high-dose group, middle-dose group and low-dose group were higher than that in the model group ((5.565±1.170、9.631±0.566、8.525±0.50 vs 3.674±0.626, all P﹤0.01). Conclusion After the radiation, the expression of P-Smad2、P-Smad3 protein were up-regulated and the expression of Smad6、Smad7 protein were down-regulated in the rats’ lung tissues. Second, the Modified Maxing Shigan Decoction can down-regulated the expression of P-Smad2、P-Smad3 protein and up-regulated the expression of Smad6、Smad7 protein in the acute radiation pneumonitis rats’ lung tissues.
