      目的:通过观察参附注射对脓毒症大鼠心肌细胞凋亡及Bcl-2、Bax蛋白表达的影响,阐明参附注射液抗脓毒症心肌损伤的相关机制。方法:雄性SD大鼠随机分为3组:假手术组、模型组和参附组,每组8只。以盲肠结扎穿孔(Cecal Ligation and Puncture,CLP)法建立脓毒症大鼠动物模型,观察各组大鼠心肌酶谱变化;凋亡原位末端标记法(TUNEL) 检测心肌细胞凋亡指数,RT-PCR 法检测心肌细胞Bcl-2 mRNA、Bax mRNA表达。结果:参附组大鼠心肌酶谱升高程度较模型组低(P<0.05),模型组较假手术组心肌酶水平高(P<0.05);参附组大鼠心肌细胞TUNEL评分平均光密度(MOD)较模型组明显降低(P<0.05),模型组较假手术组升高(P<0.05);参附组大鼠心肌细胞Bcl-2 mRNA表达较其他两组均升高(P<0.05),而Bax mRNA表达较其他两组降低(P<0.05)。结论:参附注射液能减轻脓毒症大鼠的心肌损伤,其作用机制与上调Bcl-2 mRNA的表达,下调Bax mRNA表达,减少心肌细胞凋亡相关。

      Objective:To invesgate the anti myocardial injury mechanism of Shengfu Injection by the varies of myocardial enzyme、expression of BCL-2、Bax and myocardial cell apoptosis in rat model with sepsis. Methods:24 male SD rats were randomly divided three groups: sham-operated group、model group and Shengfu group,every group had 8 rats. Rat sepsis models were established through cecal ligation and puncture,rats in model group were injected via the tail vein with 5ml/kg saline at 0h、6h、12h after making model,and at the same time point the rats in Shengfu group were injected with 5ml/kg Shengfu Injection via the tail vein,but rats in sham-operated group were only operated.24 hours after sepsis myocardial creatine kinase and it’sisozyme levels in plasma were detected,and myocardial apoptosis were detected by TUNEL(TdT dUTP nick and labelling)technique,expression level of Bcl-2 mRNA、Bax mRNA in myocardial cell were detected by RT-PCR. Results: the myocardial creatine kinase and it’sisozyme levels in model group were highest between three group,the secnod was in Shengfu group(P<0.05). The mean optical density in Shengfu group was lower than that in model group(P<0.05),and was higher than that in sham-operated group(P<0.05). Expression level of Bcl-2 mRNA in myocardial cell in model group was highest between three group(P<0.05),while Expression level of Bax mRNA in myocardial cell in model group were significant decreased (P<0.05).Conclusons:Shengfu injection could alleviate injury in cardiac muscle caused by sepsis by increasing the Bcl-2 mRNA expression and decreasing the Bax mRNA expression and reducing myocardial apoptosis.
