目的 研究高葡萄糖浓度刺激下牙周膜干细胞(PDLSCs)的增殖能力和成骨分化能力。方法 组织块法体外培养非糖尿病患者PDLSCs,分别用0mg/L(C组)、1100mg/L(L组)、4500 mg/L(H组)浓度葡萄糖刺激,MTT法检测PDLSCs增殖能力,矿化诱导后茜素红染色观察矿化结节形成,实时定量聚合酶链反应(RT-PCR)检测Runt相关转录因子2(Runx-2)、碱性磷酸酶(alkaline phosphatase,ALP)和I型胶原(collagen type I,Col-1)成骨相关基因表达。结果 MTT法检测显示H组OD值较L组和C组低(P<0.05);21天成骨诱导后,H组矿化结节面积较L组和C组少(P<0.05);成骨诱导期间,H组ALP、Runx2和Col-1诱导前后相对倍增数较L组和C组低(P<0.05)。结论 高浓度葡萄糖抑制PDLSCs增殖能力和成骨分化能力。 |

Objective:To isolate the periodontal ligament stem cells(PDLSCs) and studied the effect of high glucose concentration levels on the cellular activity and osteogenic differentiation of PDLSCs from non-diabetic patients in vitro.Methods: PDLSCs were cultured for 14 days in a low glucose medium (1100 mg/L of glucose) or in a high glucose medium (4500 mg/L of glucose),respectively, a 3-(4,5-dimethylithiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay for cellular viability was performed, the osteogenic differentiation capacity of PDLSCs was evaluated by alizarin red staining and real-time quantitative reverse transcription polymerase chain reaction(real time RT-PCR).Results:High glucose signi?cantly inhibited the proliferation of PDLSCs and reduced the optic density(OD) of the MTT, the difference was a significant(P﹤0.05), Concerning the mineralized nodule formation, the percentage of the calci?ed area and the expression of osteogenic genes (ALP, Runx-2, Col-1) to the total culture dish of PDLSCs in high glucose level were significant lower than that in the low glucose medium and control medium(P﹤0.05). Conclusion: High glucose inhibits the proliferation and differentiation of PDLSCs. |
