      目的:探讨miR-106b对乳腺癌诊断的参考意义,为诊断乳腺癌提供新的方法。方法:荧光定量PCR检测正常乳腺细胞系MCF-10A及乳腺癌细胞系MCF-7、MDA-MB-231、MDA-MB-435的miR-106b表达水平。收集30例乳腺癌患者临床病例资料,采集患者血浆,以正常人血浆作为对照,运用RT-PCR方法分析血浆miR-106b相对表达量与临床特征的关系, 评估血浆miR-106b对乳腺癌的诊断效能。 结果:MCF-10A细胞miR-106b的相对表达量为1.00±0.08,MCF-7细胞为5.15±0.45,MDA-MB-231细胞为6.86±0.57,MDA-MB-435细胞为3.86±0.29。乳腺癌患者血浆及正常人血浆中miR-106b相对表达量分别为4.13±2.47和1.00±0.75 (p<0.05)。乳腺癌患者中miR-106b相对表达量与患者年龄、肿瘤大小无显著的相关性(p>0.05),而与肿瘤的分化程度、临床分期有关(p<0.05)。经ROC曲线分析,miR-106b对乳腺癌患者和正常对照中诊断乳腺癌的AUC为0.831 (95%CI:0.719-0.942),敏感性和特异性分别为86.67%和76.67%。结论:乳腺癌患者血浆miR-106b的异常高表达可能成为乳腺癌诊断的新靶点。

      Objective: To investigate the diagnostic value of miR-106b in patients with breast cancer. Methods: The expression of miR-106b was detected by qPCR in normal breast cell line MCF-10A and breast cancer cell line MCF-7, MDA-MB-231 and MDA-MB-435. Thirsty patients with breast cancer were involved to detect the expression levels of miR-106b by RT-PCR, and then the relationship of miR-106b level and patients’ clinical features as well as the diagnostic efficiency were analyzed. Results: The relative expression levels of miR-106b in cell lines were as follows: MCF-10A (1.00±0.08), MCF-7(5.15±0.45), MDA-MB-231(6.86±0.57), MDA-MB-435(3.86±0.29). The relative expression of miR-106b in healthy control’s plasma is 1.00±0.75, and the value is 4.13±2.47 in patients’ plasma (p<0.05). In breast cancer patients, the expression levels of miR-106b had no significant correlation with age and tumor size (p>0.05), but exhibited distinct difference among patients at different tumor differentiation levels and TNM stage (p<0.05). For discriminating breast cancer patients from normal people, the area under the receiver operating characteristic curve (ROC) for plasma miR-106b was 0.831 (95%CI:0.719-0.942), and the sensitivity and specificity were 86.67% and 76.67%. Conclusion: The abnormal expression of miR-106b may become a new target in the diagnosis in breast cancer.
