| 目的：探讨中药黄芩提取物黄芩素对人胆囊癌的生物效应及机制。方法：将人胆囊癌细胞系SGC996用黄芩素处理48小时,采用MTT法检测治疗后SGC996的细胞活力；通过对透过transwell膜的SGC996细胞进行计数评估黄芩素对胆囊癌侵袭转移能力的影响；通过流式细胞术检测黄芩素处理后,SGC996细胞的凋亡情况并用western blot方法检测细胞锌指X染色体蛋白(zinc finger X-chromosomal protein,ZFX)的表达；结果：对照组,10μmol/L黄芩素组,20μmol/L黄芩素组,40μmol/L黄芩素组,80μmol/L黄芩素组,160μmol/L黄芩素组,320μmol/L黄芩素组对SGC996细胞活力的抑制率分别为0,0.4±0.9,4.2±1.5,19.3±3.8,37.9±5.8,61.6±7.8,84.2±10.2 (p<0.05)。对照组,40μmol/L黄芩素组,80μmol/L黄芩素组,160μmol/L黄芩素组对SGC996细胞的凋亡诱导率分别为0.5±0.5,7.5±1.2,16.3±1.9,31.2±2.8 (p<0.05)；对SGC996细胞转移抑制率分别为0,25.6±6.6,57.3±7.9,84.1±11.9 (p<0.05)。Western blot结果显示黄芩素对ZFX有显著的抑制作用。结论： 黄芩素有良好的抗神经胆囊癌的生物活性,其抗肿瘤效应的机制可能和下调ZFX蛋白有关。 |

| objective: To explore the effect and mechanism of baicalein extracted from Scutellaria baicalensis Georgi in gallbladder carcinoma. Methods: SGC996 cells were treated with baicalein and the cell viability was measured using the MTT assay. Cancer metastasis was evaluated by transwell assay. The apoptosis of SGC996 was measured by flow cytometry and the expression of ZFX was detected by western blot in SGC996 cells treated with baicalein. Results: The cell viability inhibitory rate of SGC996 cells in control group, 10μmol/L baicalein group, 20μmol/L baicalein group, 40μmol/L baicalein group, 80μmol/L baicalein group, 160μmol/L baicalein group, 200μmol/L baicalein group is as follows: 0,0.4±0.9,4.2±1.5,19.3±3.8,37.9±5.8,61.6±7.8,84.2±10.2 (p<0.05). The apoptosis ratio of SGC996 cells in control group, 40μmol/L baicalein group, 80μmol/L baicalein group, 160μmol/L baicalein group is as follows: 0.5±0.5,7.5±1.2,16.3±1.9,31.2±2.8 (p<0.05); Metastasis inhibitory ratio in these group is as follows: 0,25.6±6.6,57.3±7.9,84.1±11.9 (p<0.05). Western blot analysis showed the expression of ZFX was significantly inhibited by baicalein treatment in SGC996 cells. Conclusion: Baicalein showed a significant anti-tumor effect in human gallbladder cancer, the mechanism may due to the down-regulation of ZFX. |
