      目的 观察vitexicarpin对GH3垂体瘤细胞生长和增殖的影响,并探讨其作用机制。方法 通过绘制含不同浓度梯度的vitexicarpin培养基条件GH细胞生长曲线观察 Vitexicarpin对GH3细胞的作用;通过CCK-8试剂盒检测Vitexicarpin作用后的剂量时间效应,应用流式细胞检测细胞凋亡率以及Western Blotting测定 PARP, Caspase-3等凋亡相关蛋白表达的方法探索Vitexicarpin的作用机制。 结果 vitexicarpin可以抑制GH3细胞的生长和增殖,这种抑制作用有明显的剂量和时间效应,流式细胞检测分析结果显示其抑制增殖作用是通过诱导GH3细胞凋亡实现的,Western Blotting检测发现Caspase-3活化,PARP裂解。结论 Vitexicarpin可以抑制GH3垂体腺瘤细胞增殖,其可能机制是通过线粒体介导的细胞凋亡通路。
英文摘要:
      Objectives To investigate the anti-proliferration effect on pituitary GH3 cells and explore the molecular mechanism. Methods We evaluate the proliferation of GH3 cells treated by different solubility gradient of vitexicarpin by drawing the cell viability curve. We used CCK-8 method to evaluate the time- and dose-dependent effects. The effect of vitexicarpin on GH3 cell apoptotic and the change of apoptotic protein PARP, Caspase 3 were evaluated by using flow cytometer and western blot respectively. Therefore, the molecular mechanism of vitexicarpin on GH3 cell was clarified. Results Vitexicarpin can inhibit the growth and proliferation of GH3 cells and this inhibitory is a time-and dose-dependent effect, the results of flow cytometry showed that v itexicarpin can promote GH3 cell apoptosis. Treated by vitexicarpin caspase-3 was activeded and PARP was degradated. Conlusions We concluded that v itexicarpin can inhibit proliferation of GH3 cell and mitochondria dependent apoptosis may partly be the mechanism.
