| 目的：探讨水飞蓟宾联合顺铂对人肺癌细胞A549的杀伤效应及机制。方法：将人肺癌细胞A549分别用水飞蓟宾,顺铂及两者联合治疗后,采用MTT法检测不同治疗方案对A549细胞的抑制作用,通过流式细胞术检测细胞的凋亡,通过western blot法检测细胞Bcl-2与Bax的表达；利用小RNA抑制Bax的表达后,检测水飞蓟宾联合顺铂对A549细胞凋亡的诱导能力。结果：水飞蓟宾联合顺铂可显著抑制A549细胞的增殖并诱导其发生凋亡。水飞蓟宾联合顺铂诱导A549细胞凋亡依赖于细胞内Bcl-2表达水平的降低和Bax表达水平的提高。结论：水飞蓟宾联合顺铂通过降低Bcl-2/Bax比例诱导人肺癌细胞发生凋亡。 |

| objective: To explore anticancer activity and mechanism of silibinin combine with cisplatin in A549 lung cancer cells. Methods: A549 cells were treated with silibinin, cisplatin and both. Cell proliferation was measured using the MTT assay. Induction of apoptosis was determined by flow cytometry. The expression of Bcl-2 and Bax was detected by western blot. The expression of Bax was inhibited by siRNA, and then the apoptosis of A549 was measured after treated with silibinin combine with cisplatin. Results: Silibinin combine with cisplatin significantly inhibited the proliferation and induced apoptosis in A549 cells. The expression of Bcl-2 significantly decreased but the Bax highly increased in A549 after treated with silibinin combine with cisplatin. Conclusion: Silibinin combine with cisplatin induced apoptosis of A549 cells via decreasing the Bcl-2/Bax ratio. |
