      目的:通过对哮喘小鼠肺组织水通道蛋白气道黏蛋白(MUC)5ac mRNA的影响,探讨麻杏二三汤抑制哮喘小鼠气道黏液高分泌的机制。方法:雌性BALB/c小鼠60只,随机分成①空白对照组、②哮喘模型组、③强的松组、④中药(麻杏二三汤)高剂量组、⑤中药中剂量组、⑥中药低剂量组。以腹腔注射0.1%卵白蛋白(OVA)与10%氢氧化铝混悬溶液致敏,以1%OVA雾化液雾化攻击制作哮喘小鼠模型。①~⑥组分别给予生理盐水、生理盐水、强的松(10.0mg/kg)、麻杏二三汤高剂量(30g/kg)、中剂量(15g/kg)、低剂量(7.5g/kg)灌胃,Real-time PCR测定各组MUC5ac mRNA表达。结果:麻杏二三汤高、中、低剂量组△Ct值均高于哮喘模型组(P<0.01)。结论:麻杏二三汤能够有效抑制哮喘小鼠气道黏液高分泌的机制可能与其能够下调支气管肺组织中MUC5ac mRNA的表达有关。
英文摘要:
      Objective Through experiments to study the effect of Maxingersan decoction to the asthmatic mice’ lung MUC5ac mRNA,to discuss the relationship between the MUC5ac mRNA and the phlegm of traditional chinese medicine and the mechanism of Maxingersan decoction inhibiting the asthmatic mice’ airway mucus hypersecretion .Methods Taking 60Sfemale BALB/cSmice which weight are between 18 and 22 grams randomly divide into 6 groups, ①the blankScontrol group, ②the asthmaSmodelSgroup, ③the dexamethasone group,④the Maxingersan decoction high dose group,⑤the Maxingersan decoction medium dose group,⑥the Maxingersan decoction low dose group. IntraperitonealSinjection by the fresh mixture of 0.1%ovalbumin(OVA)and 10% Aluminum hydroxide suspension solution to make the mice sensitization, and atomizing with 1% OVA atomized liquid to induce asthma. The first to the sixth group are respctively gavaged by NS, NS, prednisone (10.0mg/kg), Maxingersan decoction(7.5g/kg), Maxingersan decoction(15g/kg), Maxingersan decoction (30g/kg). The Real-timeS fluorescenceSquantitativeSpolymerase chain reaction assay (RT-PCR) to o bserve the change of MUC5ac mRNA in the mice’ lung tissue of each group. Results The △Ct numerus of all the Maxingersan decoction groups are higher than theSasthmaSmodelSgroup (P<0.01 or (P<0.05). Conclusion The mechanism of Maxingersan decoction can effectively inhibit the asthmatic mice’ airway mucus hypersecretion may be related to it can downregulate theSexpressionSofS MUC5ac mRNA.
