      目的:探讨炙马钱子对实验性自身免疫性重症肌无力(EAMG)大鼠的免疫调节机理。方法:采用鼠源乙酰胆碱受体α亚基97-116肽段方法免疫Lewis大鼠建立实验性重症肌无力模型,观察实验大鼠的临床表现,并将建模成功的EAMG大鼠随机分为模型组,强的松组,炙马钱子低剂量、中剂量和高剂量组,采用酶联免疫吸附试验(ELISA)法检测正常对照组和成模各组大鼠血清中体液免疫指标AChRab与细胞免疫指标IL-6的含量情况并比较各组之间的差异。结果:大鼠EAMG模型组血清中AChRab和IL-6含量相比正常组显著升高(P<0.01),药物干预后各组含量都有所下降。炙马钱子高剂量组AChRab含量比中、低剂量组下降更显著(P<0.01),与强的松组比无显著差异(P>0.05);炙马钱子各剂量组IL-6含量均低于强的松组,特别是中、高剂量组更明显低于强的松组,差异有统计学意义(P<0.01);炙马钱子中、高剂量组之间的差异无统计学意义(P>0.05)。结论:炙马钱子能降低EAMG大鼠血清中AChRab和IL-6含量,抑制T、B细胞活化,减轻对突触后膜AchR的损害,缓解EAMG病情,一定剂量范围内的炙马钱子在免疫调节方面表现优于强的松。

      Objective:To investigate the immune regulation mechanism of nux vomica on experimental autoimmune myasthenia gravis (EAMG). Method: Lewis rats were immunized with Torpedo-derived 97-116 Peptide of the AchR α-subunit in CFA. After the second immunization, the rats"weights and symptoms were observed to judgement the EAMG model were succeed made.Then these succeed made EAMG model were divided into the model group,the prednisone group,the low dosage、the middle dosage and the high dosage of nux vomica groups,administered by Prednisone and nux vomica intervention treatment. Observed the effect of each group to the clinical feature and examined the level of AchRab and IL-6 by using the way of enzyme linked immunosorbent assay(ELISA). Results:Compared to the normal group,the level of AChRab of high dosage of nux vomica was remarkablely decrease than the middle and low dosage of nux vomica(P<0.01),but has no difference to prednisone group(P>0.05).The level of IL-6 of each group of nux vomica was significantly lower than prednisone group(P<0.01),especially the high and middle dosage of nux vomica.But there is no difference between high dosage and middle dosage(P>0.05). Conclusion:Nux vomica can reduce the level of AChRab and IL-6 in EAMG model, inhibit the activation of T cell and B cell, so that it can reduce the damage of postsynaptic membrane AchR, alleviate the development of EAMG.The effect of nux vomica was better than prednisone in improving immune adjustment in certain dosage.
