| 目的:研究大肠癌围手术期气虚证与阴虚证患者的特征性基因差异表达谱。方法: 选择大肠癌围手术期的主要中医证候气虚证，分别以正常组作阴性对照和阴虚证作阳性对照，采集符合条件病例的大肠癌组织及瘤周组织，采用含有45033个己知基因的人全基因组芯片技术检测大肠组织内差异基因表达情况，比较气虚证组与阴虚证瘤体组的差异表达基因，利用生物分子功能注释系统GO和pathway对相关基因进行生物信息学分析；用实时荧光定量PCR技术验证部分差异表达基因结果的可靠性。结果：得出大肠癌围手术期气虚证瘤体组和瘤周正常组差异表达基因共5044条，其中，表达水平上调3400条；下调的有1644条；气虚证与阴虚证瘤体组差异表达基因共1335条，其中，表达水平上调547条，下调的有788条。经检索互联网生物学公共数据库GeneBank，获得每条基因的名称和其他基本信息。结论：大肠癌围手术期气虚组与瘤周正常组相比较，存在组织基因差异表达特征图谱，主要是与细胞凋亡，细胞周期，血管平滑肌收缩，嘌呤嘧啶代谢，细胞信号传导，DNA 复制等基因相关；气虚证与阴虚证瘤体组比较，也存在组织基因差异表达特征图谱，主要是与炎症、免疫应答、矿物质吸收、细胞内吞、细胞粘附、遗传信息过程、肌动蛋白细胞骨架的调控，氮代谢，细胞信号传导等相关基因。 |

| Objective:Comparison of differentially expressed genes between deficiency of Qi deficiency and Yin deficiency patients in perioperative period of colorectal carcinoma patient.Methods:we mainly focus on Qi deficiency and Yin deficiency syndrome in perioperative period of colorectal carcinoma patients,and respectively set up a normal group as a negative control group,gathering the colorectal tumor tissue and around tissue which are fit requirement.Test the tissue differently expression of gene by human gene technology which with 45033 known human Gene.After analyzing the gene chip image of qi deficiency group and Yin deficiency group.Pathway and GO analysis were applied to analyze the related gene biological information.Finally,some differentially expressed genes in different groups are verified through real-time PCR.Results:We found that a total of 5044 genes expressed differentially in colorectal tissue between Qi deficiency group and normal control group,among which 3400 were up-regulated and 1644 were down-regulated.After retrieval the public databank,we knew names and basic information of all the genes.we knew the differentially expressed genes between Qi deficiency syndrome and normal group were involved in:apoptosis,cell cycle,DNA replication, purine and Pyrimidine metabolism,signaling pathway, vascular smooth muscle contraction and so on.And a total of 1335 genes expressed differentially in colorectal tissue between Qi deficiency group and normal control group,among which 547 were up-regulated and 788 were down-regulated.After retrieval the public databank,we knew names and basic information of all the genes.Conclusions:Comparing with two groups, differentially expressed genes map as follow:inflammation,Immune response,mineral absorption,Endocytosis, signaling pathway and so on. |
