      目的:探讨刀豆素A对人乳腺癌细胞MCF-7的杀伤作用及其机制。方法:采用MTT法检测不同浓度ConA(concanavalin A) 处理MCF-7细胞后,MCF-7的细胞增殖情况并检测自噬标记蛋白LC-3Ⅱ,加入自噬特异性抑制剂3-甲基腺嘌呤(3-MA)后再检测MCF-7细胞对ConA的敏感性。利用基因沉默技术,抑制BNIP3的表达,再检测ConA处理后MCF-7细胞的增殖和自体吞噬。结果:ConA可显著抑制MCF-7细胞的增殖并诱导其发生自噬性死亡。ConA处理后MCF-7细胞的BNIP3显著增高,利用小RNA抑制BNIP3的表达后,ConA对MCF-7的增殖抑制作用降低并抑制了ConA诱导的自体吞噬效应。结论:刀豆素A通过上调BNIP3的表达诱导人乳腺癌细胞发生自噬性死亡

      objective: To observe the proliferation of MCF-7 cells treated with various concentrations of concanavalin A and the expression of Autophagy marker protein-LC-3Ⅱ. Methods: Using gene silencing technology to suppress the expression of BNIP3, and then detected the proliferation and autophagy of MCF-7 treated with ConA. Adding 3-MA and then detect the proliferation of MCF treated with ConA. Results: ConA significantly inhibited the proliferation of MCF-7 cells inducing autophagic cell death. Expression of BNIP3 significantly increased in MCF-7 when treated with ConA. Although, inhibited the expression of BNIP3 using small RNA, the effect of suppression as well as autophagy of ConA decreased in MCF-7. Conclusion: concanavalin A induced autophagic cell death of MCF-7 cells via upregulating the expression of BNIP3.
