目的:探讨三七总皂甙(Panaxnonginsengsaponins, PNS)对大鼠肝缺血再灌注损伤的保护作用,并研究其对氧化应激及炎症反应相关因子的影响。 方法:30只健康雄性SD大鼠,体重250-300g,随机分为三组:假手术组(Sham组),肝缺血再灌注组(HIR组)和三七总皂甙治疗组(PNS组)。PNS组术前1h腹腔注射给予50mg/kg的PNS生理盐水溶液(浓度为5mg/mL),Sham组及HIR组则均于术前1小时给予相同剂量的生理盐水。预处理3小时后处死,比较评估三组肝组织MDA、SOD、MPO、ICAM-1及血清TNF-α、IL-1β变化。 结果: 应用三七总皂苷后,肝缺血再灌注损伤后肝组织中MDA水平、MPO活性、ICAM-1含量及血清TNF-α、IL-1β明显下降(P<0.05),而SOD活性则有明显上升(P<0.05)。 结论: 三七总皂甙可以通过减少氧化应激水平以及炎症因子表达进而减轻肝缺血再灌注损伤,起肝脏保护作用。 |

Objective: To explore the protective effect of Panaxnonginsengsaponins (PNS) on hepatic ischemia reperfusion injury (HIR) in rat, and find possible relationship to oxidative stress and inflammatory factors. Methods: 30 SD rats were divided to three groups, as Sham group, HIR group, and PNS treatment group. The rats in PNS group were given 50mg/kg PNS solution (5mg/ml) by intraperitoneal injection at 1h before surgery, while other groups accept similar dose of saline. After 3 hours from pre-injection, all rats were sacrificed, and liver MDA, SOD, MPO, ICAM-1 and serum TNF-α、IL-1β were tested by ELISA kit. Results: The appliance of PNS gave rise to decreases in liver MDA, MPO, ICAM-1, and serum TNF-α, IL-1β, while SOD increased (P<0.05). Conclusion: PNS can protect liver after HIR by deceasing oxidative injury and inflammatory factors express. |
