      目的:研究PNS对人急性单核细胞白血病细胞系U937细胞的增殖、凋亡及Caspase-3表达的影响。方法:用不同浓度的PNS作用于U937细胞,采用MTT法检测细胞增殖活性,用流式细胞术检测细胞周期和凋亡,western blot法检测Caspase-3蛋白表达。结果:PNS抑制U943细胞生长并呈时间和剂量依赖性;100、200、400、800 mg/L处理U943细胞24h,细胞凋亡率分别为5.930.15%、7.430.70%、12.30.38%和20.40.91%,对照组细胞凋亡率为(1.731.50%)(P<0.01),并诱导细胞G1期阻滞;western blot结果显示PNS使Caspase-3蛋白表达上调。结论:PNS能抑制U937细胞增殖,并可能通过上调Caspase-3蛋白表达诱导细胞凋亡。
英文摘要:
      objective: To investigate the effect of PNS on proliferation, apoptosis and expression of caspase-3 of human acute monocytic leukemia cell line U937. Methods: U937 cells were treated with PNS an different concentration. Cell proliferation was analyzed by MTT assay. Cell cycle and apoptosis by flow cytometry and Caspase-3 protein expression by western blot. Results: The inhibition of cell proliferation in U937 cells exposed to PNS was dose and time dependent manner. 24 hours after exposure to 100, 200, 400, 800mg/L PNS, the apoptotic rates of U937 were 5.930.15%, 7.430.70%, 12.30.38% and 20.40.91%, respectively, which were markedly higher than that of control(1.731.50%)(P<0.01), and include the G1 phase arrest of U937 cells. The expression of Caspase-3 protein was up-regulated. Conclusion: PNS can inhibit the proliferation of U937 cells, and induce apoptosis by up-regulating the expression of Caspase-3.
