      目的:观察姜黄素对肾缺血再灌注损伤及缺氧诱导因子-1α(HIF-1α)表达的影响。方法: SD大鼠30只,随机分为假手术组、缺血再灌注组和姜黄素组,每组10只。姜黄素组术前连续3天 腹腔注射姜黄素200mg/kg,1天1次;缺血再灌注组术前3天则腹腔注射等容量生理盐水,1天1次, 假手术组模拟手术操作过程但不夹闭左肾动脉,另二组均予夹闭左肾动脉,并切除右肾。左肾夹 闭45min后松开动脉夹,2h后切除左肾置10%中性甲醛液固定,进行切片HE染色及免疫组化检测 各组肾组织HIF-1α表达水平;取左肾后立即通过下腔静脉采血,检测Scr、BUN含量。结果:与缺血 再灌注组比较,姜黄素组肾脏病理损伤明显减轻,血Scr、BUN水平及HIF-1α表达显著降低,差异有 统计学意义(P<0.05)。病理结果显示,缺血再灌注组有不同程度肾小球、肾间质充血,肾小管上皮 细胞水肿、细胞内玻璃样变及脂肪变性,肾小管腔内蛋白管型明显;姜黄素组仅个别标本存在蛋白 管型,水肿减轻。结论:姜黄素对肾缺血再灌注损伤具有干预作用,可能通过调节HIF-1α因子表 达,而发挥肾脏保护作用。

      Objective: To investigate the effects of curcumin(Cur) on renal ischemia-reperfusion injury(I/R) and the expression of hypoxia-inducible factor-1 Alpha(HIF-1α) in rats. Method: Thirty Sprague-Dawley rats were randomly divided into 3 groups, including a sham operation group, renal I/R group, and I/R+cur group. The I/R and I/R+cur groups were subjected to left renal ischemia for 45 minutes and reperfusion for 2 h. The sham oper? ation group underwent a surgical procedure that included isolation of the left renal artery without occlusion. In cur treated group, the rats received intraperitoneal curcumin administration of 200 mg/kg for 3 consecutive days. The sham operation group received equivalent volume of saline treatment for the same time. The left kidneys ere taken out after 2-h reperfusion for measuring HIF-1α in renal tissue and serum specimens were immediately col? lected from inferior vena cave for measuring creatinine and blood urea nitrogen(BUN). Results: Cur pretreatment markedly reduced I/R-induced renal injury as indicated by histological alterations and the decreases in creati? nine, BUN and HIF-1α (P<0.05). HE staining showed that hyperemia in glomcrulus and renal interstitium,edema in renal tubular epithelial cells,hyalinization and steatosis in cells, and obvious protein cast in renal tubules in I/ R group, while that rare protein cast and the abatement of cellular edema in cur-treated group. Conclusion: Cur may have a protective effect in renal injury induced by I/R. The protective effect of cur may be attributed to the suppression of HIF-1α.
