      目的: 观察地骨皮水提液对炎症环境下脂肪细胞葡萄糖代谢及PPARγmRNA表达的影 响, 探讨其改善炎性脂肪细胞糖代谢的可能机制。方法: 葡萄糖氧化酶法检测地骨皮水提液联 合脂多糖(LPS)干预 72h 后 3T3-L1 脂肪细胞对培养液中的葡萄糖消耗量, RT-PCR 法检测 PPARγmRNA的表达。结果: LPS能显著降低脂肪细胞葡萄糖消耗量 (P<0.01); 2.5mg/mL地骨皮水 提液能改善LPS干预后的脂肪细胞的葡萄糖利用 (P<0.01)。LPS能显著下调 PPARγmRNA表达 (P<0.05); 2.5mg/mL、 1.0mg/mL地骨皮水提液能提高LPS干预后的脂肪细胞的PPARγmRNA表达 (P<0.05)。结论: 地骨皮水提液能改善LPS抑制的脂肪细胞的葡萄糖代谢及PPARγmRNA表达, 其 机制可能通过上调PPARγ表达来抑制炎症, 改善脂肪细胞葡萄糖代谢。
英文摘要:
      Objective: To observe the impact of the Digupi extract on glucose metabolism and the expression of PPARγ mRNA of fat cells in the inflammatory environment and to explore its possible mechanism. Methods: Glucose consumption by 3T3-L1 fat cells in the culture medium intervened with the Digupi extract and Lli? popolysaccharide for 72h was measured with the glucose oxidase method. The expression of PPARγ mRNA after intervention was detected by using RT-PCR. Results: Lipopolysaccharide significantly reduced the glucose con? sumption of fat cells (P<0.01); the Digupi extract of 2.5 mg/ml increased the glucose utilization reduced by lipo? polysaccharide (P<0.01). Lipopolysaccharide significantly lowered the expression of PPARγ mRNA (P<0.05) and the reduction was reversed by the Digupi extract of both 2.5 and 1.0 mg/ml intervention (P<0.05). Conclusion: Digupi extracts can improve the lipopolysaccharide-impaired glucose metabolism of fat cells by elevating the ex? pression of PPARγ mRNA to inhibit inflammation.
