      目的: 观察姜黄素对Lewis 肺癌小鼠瘤体组织TGF-β1、 Smad3、 Smad7蛋白表达的影响。 方法: 雄性C57BL/6小鼠20只, 随机分成姜黄素处理组与模型组, 每组10只。采用皮下接种瘤细胞 悬液造模。造模24h后, 模型组与姜黄素组分别腹腔注射注射用水和200mg/ (kg·d) 浓度的姜黄素 混悬液, 0.5mL/只, 1天1次, 连续12天。应用免疫组织化学显色技术, 检测小鼠瘤体组织内TGF-β 1、 Smad3、 Smad7蛋白的表达及定位情况。结果: ①与模型组比较, 姜黄素抑瘤率29.96%。姜黄素 组瘤体平均质量 (1.64±0.32) g, 与模型组 (2.34±0.70) g比较, 差异具有统计学意义 (P<0.05)。②本 研究采用免疫组化定量公式计算蛋白表达阳性单位, 结果显示TGF-β1染色强度姜黄素组为 (17.83±3.46), 模型组为 (25.76±4.72), 两组比较差异具有统计学意义 (P<0.01); Smad3染色强度姜 黄素组为 (14.64±2.24), 模型组为 (23.71±3.07), 两组比较差异有统计学意义 (P<0.01); Smad7染色 强度结果显示姜黄素组为 (10.97±1.89), 模型组为 (8.46±1.04), 两者比较差异具有统计学意义 (P< 0.01)。③瘤体组织内TGF-β1与Smad3蛋白表达具有相关性 (P=0.043)。结论: 姜黄素能抑制肿瘤 生长, 其作用机制可能与上调Smad7蛋白表达, 下调Smad3蛋白表达, 从而减少TGF-β1的过度表 达有关。

      Objective: To observe the effects of curcumin on the expression of TGF-β1, Smad3, Smad7 proteins in mice with Lewis lung cancer. Methods: Twenty male C57BL/6 mice were randomly divided into model group and curcumin group, 10 in each. A mouse model of Lewis lung cancer was established by subcutaneously inocu? lating tumor cell suspension. Mice in the model group were intraperitoneally injected with 0.5 ml of saline once a day and 200 mg/kg/d of curcumin suspension was injected once a day in the curcumin group, both for 12 days. Tumor mass was harvested on the thirteenth day after 12 hour fasting. The expression of TGF-β1, Smad3, Smad7 proteins in tumor tissues were assessed by immunohistochemistry. Results: ①Curcumin inhibition rate was 29.96%. The average weight of subcutaneous tumor in the curcumin group was 1.64 ±0.32 g, significantly lighter than that of the model group (2.34±0.70 g), P<0.05.②The staining intensities of TGF-β1 and Smad3 in the curcumin group were 17.83±3.46 and 14.64±2.24 as compared to 25.76±4.72 and 23.71±3.07 in the model group(P<0.01) . The staining intensity of Smad7 was 10.97±1.89 in the curcumin group, while 8.46±1.04 in the model group, with a significant difference between them(P<0.01) . ③The expressions of TGF-β1 and Smad3 pro? teins in the tumor tissue was correlated(P=0.043) . Conclusion: Curcumin can inhibit tumor growth by reducing the excessive expression of TGF-β1 as a result of upregulating Smad7 and downregulating Smad3.
