目的: 观察小白菊内酯 (parthenolide, PTN) 对人肾小球系膜细胞炎症因子表达的抑制作
用。方法: ①以MTS方法检测PTN和厚朴酚 (0~50μmol/L) 在人肾小球系膜细胞中的安全浓度。②
将系膜细胞与高糖 (50mmol/L) 及不同浓度小白菊内酯 (0~50μmol/L) 共同培养24h, 24h后ELISA法
检测培养上清液中TNF-α、 TGF-β1、 MIP-1α与MCP-1蛋白的表达水平。结果:①0~50μmol/L的
PTN浓度未对系膜细胞的增殖活力产生影响, 但更高浓度的PTN可明显降低系膜细胞的增殖活
力。②50mmol/L高糖可显著增加炎症因子TNF-α、 TGF-β1、 MIP-1α及MCP-1的蛋白水平。PTN
呈剂量依赖性地抑制高糖刺激的炎症反应作用。结论: PTN能有效抑制高糖刺激后人肾小球系膜
细胞炎症因子的表达, 从而削弱高糖所致的系膜细胞炎症反应, 为进一步阐明PTN改善DN的作用
机制提供了实验依据。 |

Objective: To observe the inhibitory effect of parthenolide (PTN)on the expression of proinflammatory
cytokines in high glucose-stimulated human mesangial cells. Methods: The safe windows of PTN and magnolol
were determined by the MTS assay. Human masengial cells were cultured with 50mmol/L glucose and 0-50μmol/
L PTN for 24 h, and then the expressions of TNF-α, TGF-β1, MIP-1α, and MCP-1 proteins in the supernatant
were measured by ELISA. Results: The safe window of PTN was 0-50μmol/L. Beyond this window, the viability
of human mesangial cells was decreased. High glucose(50mmol/L)stimulation markedly upregulated the expres?
sions of TNF-α, TGF-β1, MIP-1α, and MCP-1 proteins. PTN inhibited the high glucose-induced inflammatory re?
sponse in a dose-dependent manner. Conclusion: PTN can effectively attenuate inflammatory response of high
glucose-induced human mesangial cells by downregulating the expressions of proinflammatory cytokines. This may
be of help to understand the underlying mechanism of PTN improving diabetes nephropathy. |
