      目的: 探讨经皮外周血管腔内成形术 (PTA) 后活血解毒益气方药物血清对人脐静脉内皮 细胞 (HUVECs) 分泌ICAM-1、 MCP-1的影响。方法: 新西兔35只, 随机分为正常组、 模型组、 普伐 他汀组及活血解毒益气方大、 小剂量组, 各7只。除正常组外, 另四组首先运用球囊原位扩张、 拉伤 致兔颈总动脉PTA术后内皮损伤模型。术前3天各组予相应药物给药, 造模成功后继续给药15 天, 获取模型动物和给药动物血清; 采用ELISA法观察药物血清作用24h和48h, 对人脐静脉内皮细 胞分泌ICAM-1、 MCP-1活性的影响。结果: 活血解毒益气方大、 小剂量组和普伐他汀组药物血清 作用24h均能显著抑制HUVECs分泌ICAM-1, 与模型组比较, P<0.05; 血清作用48h, 普伐他汀组、 大小剂量组仍具有显著的抑制作用,P<0.05。各组药物血清作用24h、 48h均能显著降低HUVECs 分泌MCP-1, 与模型组比较差异有统计学意义, P<0.05。结论: 活血解毒益气中药可通过抑制HU? VECs分泌ICAM-1、 MCP-1, 抑制炎症反应。
英文摘要:
      Objective: To evaluate the effect of Huoxuejieduyiqifang on the expression of ICAM-1 and MCP-1 of human umbilical vein endothelial cells (HUVECs) . Methods: Thirty-five New Zealand rabbits were randomly di? vided into normal control group,model group,pravastatin group,and high-and low-dose Huoxuejieduyiqifang groups, 7 in each. Endothelial injury was established by percutaneous transluminal coronary angioplasty (PTCA)in other 4 groups except the normal control group. To obtain the mediated serum, the pravastain and Huoxuejieduyiq? ifang groups were mediated with the corresponding drugs for 3 days before PTCA and 15 days after PTCA, and the normal control and model groups were given equivalent saline for the same period. Then HUVECs were cul? tured with the mediated sera, respectively, and the expressions of ICAM-1 and MCP-1 were assessed by ELISA in 24 h and 48 h after intervention. Results: Compared with the model group, the expression of ICAM-1 in the pravastatin and high- and low-dose Huoxuejieduyiqifang groups was significantly decreased in 24 h and 48 h af? ter intervention (both P<0.05) . Conclusion: Huoxuejieduyiqifang can attenuate inflammation by inhibiting the ex? pression of ICAM-1 and MCP-1 of HUVECs.
