      目的:探讨雷公藤多苷对佐剂性关节炎(AA)大鼠P 物质(SP)的干预作用。方法:以AA 大 鼠为研究对象,以肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)作为炎症活动的指标,造模成功 后,予雷公藤多苷片混悬液灌胃治疗30 天,分别在治疗前后用放射免疫法测定各组大鼠血液、滑 膜中的TNF-α、IL-1β、SP 以及脊髓后角中的SP 水平, 以观察雷公藤多苷治疗后各组织SP 的变 化。结果:与正常组比较,模型组大鼠血液和滑膜中的SP、TNF-α、IL-1β 和脊髓后角中的SP 水平 明显增高(P<0.05); 与模型组比较, 给药30 天后, 给药组大鼠的右足趾肿胀较模型组减轻(P< 0.05),给药组大鼠血液、滑膜中TNF-α、SP 及滑膜中IL-1β 水平都显著降低(P<0.05),而脊髓后角 中SP水平未见明显下降(P>0.05)。结论:雷公藤多苷具有抗炎镇痛免疫抑制作用,其作用机制除降 低血清和滑膜中的TNF-α、IL-1β 水平外, 还可能与降低佐剂性关节炎大鼠外周的SP 水平有关, 但对中枢SP无影响。
英文摘要:
      Objective:To investigate the effects of tripterygium polyglucoside on substance P(SP)in adjuvant arthritis( AA) rats. Methods: AA rats were randomly divided into the normal control group,the model group, and the treatment group. Tumor necrosis factor-alpha(TNF-α)and interleukin-1beta(IL-1β)were used as indicators of inflammatory activity. The treatment group was intragastrically administered with tripterygium polyglucoside for 30 days after modeling. The levels of TNF-α,IL-1β,SP in peripheral sera and the synovial membrane and the level of SP in the spinal dorsal horn were measured by radioimmunoassay before and after treatment. SP level was compared among groups. Results: In the model group, the levels of SP in the spinal dorsal horn and TNF-α,IL-1β, SP in the blood and synovial membrane increased compared with those in the normal control group(P<0.05). After treatment with tripterygium polyglucoside,the swelling of the right toe was attenuated(P<0.05), and the levels of SP, TNF-α in the blood and synovial membrane and IL-1βin the synovial membrane were all lower in the treatment group,as compared with those in the model group(P<0.05). However, the level of SP in the spinal dorsal horn was not significantly different in the 2 groups after treatment(P>0.05).Conclusion:Tripterygium polyglucoside in the treatment of AA rats had favorable anti-inflammatory and analgesic effects that may involve reducing the levels of the peripheral SP besides TNF-α and IL-1β in the blood and synovial membrane, but had no effect on central SP.
